چکیده

Recent evidence has revealed that small polypeptides (containing fewer than 100 amino acids) can be translated from noncoding RNAs (ncRNAs), which are usually defined as RNA molecules do not encode proteins. However, studies on functional products primary transcripts of microRNA (pri-miRNA) quite limited. Here, we describe a peptide termed miPEP31 is encoded by primiRNA-31. highly expressed in Foxp3+ regulatory T cells (Tregs) and significantly promotes the differentiation Tregs without affecting their inhibitory ability. Our results show cell-penetrating both vitro vivo. downregulates miR-31 expression, enhances peripheral Treg induction, dramatically suppresses experimental autoimmune encephalomyelitis. Mechanistically, acts transcriptional repressor inhibiting expression miRNA-31, negative regulator Tregs. reveal an indispensable role maintaining immune homeostasis promoting also present potential therapeutic for modulating miRNA treating diseases.

Cyclic AMP-Responsive Element-Binding Protein (CREB) is Critical in Autoimmunity by Promoting Th17 but Inhibiting Treg Cell Differentiation

The molecular mechanisms that govern differential T cell development into pro-inflammatory Th17 vs. regulatory T (Treg) cells remain unclear. Here, we show that selective deletion of CREB in T cells or Th17 cells impaired Th17 cell differentiation in vitro and in vivo, and led to resistance to autoimmune diseases. Mechanistically, CREB, activated by CD3-PKC-ϴ signaling, plays a key role in regu...

Targeting pre-miRNA by Peptide Nucleic Acids

PNAs conjugated to carrier peptides have been employed for the targeting of miRNA precursor, with the aim to develop molecules able to interfere in the pre-miRNA processing. The capability of the molecules to bind pre-miRNA has been tested in vitro by fluorescence assayes on Thiazole Orange labeled molecules and in vivo, in K562 cells, evaluating the amount of miRNA produced after treatment of ...

Regulation of pri-miRNA processing by a long noncoding RNA transcribed from an ultraconserved region.

Noncoding RNAs (ncRNAs) control cellular programs by affecting protein-coding genes, but evidence increasingly points to their involvement in a network of ncRNA-ncRNA interactions. Here, we show that a long ncRNA, Uc.283+A, controls pri-miRNA processing. Regulation requires complementarity between the lower stem region of the pri-miR-195 transcript and an ultraconserved sequence in Uc.283+A, wh...

Neutron-encoded protein quantification by peptide carbamylation.

We describe a chemical tag for duplex proteome quantification using neutron encoding (NeuCode). The method utilizes the straightforward, efficient, and inexpensive carbamylation reaction. We demonstrate the utility of NeuCode carbamylation by accurately measuring quantitative ratios from tagged yeast lysates mixed in known ratios and by applying this method to quantify differential protein expr...

A 5′-uridine amplifies miRNA/miRNA* asymmetry in Drosophila by promoting RNA-induced silencing complex formation

BACKGROUND MicroRNA (miRNA) are diverse in sequence and have a single known sequence bias: they tend to start with uridine (U). RESULTS Our analyses of fly, worm and mouse miRNA sequence data reveal that the 5'-U is recognized after miRNA production. Only one of the two strands can be assembled into Argonaute protein from a single miRNA/miRNA* molecule: in fly embryo lysate, a 5'-U promotes m...
